Virtual Screening
FieldScreen is a ligand-based virtual screening system that returns a set of novel bioisosteres with or without knowledge of the target’s protein structure. In this project, FieldScreen was used to discover potential lead structures that could inhibit 11β-hydroxysteroid dehydrogenase type 1, which may be involved in the control of obesity, insulin resistant diabetes and cognition. The project resulted in 408 purchased structures and several patent filings for lead candidates. Read the full case study here.
Library Design
Identifying a maximum number of potentially active chemical scaffolds while ensuring the resulting compound library is tractable and cost-effective for routine screening is extremely important in library design. In this case study, FieldTemplater was used to select a diverse library of potential H3 antagonists and the results include many novel active compounds, which couldn’t have been developed without the expert use of FieldTemplater software. SAR Analysis & Lead Generation
This project explored the structure activity relationships (SAR) of compounds that have potential in the fight against chronic obstructive pulmonary disease (COPD), a progressive disease prevalent in industrialized countries. The analysis was used in the selection of compounds for use in virtual screening and fragment replacement experiments in order to generate new leads. The experiments resulted in the discovery of novel chemotypes and lead candidates. Read the full case study here.
Scaffold Hopping
One of the best ways to discover new intellectual property is to perform scaffold hopping on known active compounds. In this case study, we used FieldStere to find novel structures that mimic a known allosteric antagonist (MPEP) of a metabotropic glutamate receptor. The search results contain both known actives and previously unreported active compounds, providing an excellent starting point for developing new intellectual property.
